白花蛇舌草总黄酮诱导人胃癌BGC-823细胞凋亡及其作用机制

摘要
目的:研究白花蛇舌草总黄酮(FOD)对体外培养的人胃癌BGC-823细胞增殖、凋亡的影响及其可能的分子机制。方法:1.相差显微镜观察梯度浓度FOD作用BGC-823细胞48h细胞形态变化。2.MTT法测定FOD对细胞增殖能力的影响。3.流式细胞术(FCM)测定药物作用后细胞周期和凋亡率的改变。4.Western-blot法评价药物作用48h后细胞埃茲蛋白(Ezrin)、细胞间黏附分子1(ICAM-1)的表达情况。结果:1.与阴性对照组相比FOD可使BGC-823细胞连接减少;细胞体积缩小,细胞核固缩,染色质边集。2.FOD作用细胞24h、48 h后半数抑制浓度(IC50)依次为12.58μg/ml、8.43μg/ml。3.FOD使BGC-823细胞呈G0/G1细胞周期阻滞(p<0.05),并诱导细胞凋亡(p<0.05)。4.不同浓度药物作用细胞48h后Ezrin表达量下降而ICAM-1表达量升高。结论:1.FOD可抑制人胃低分化腺癌BGC-823细胞增殖,且与药物作用时间和剂浓度呈正相关。2.FOD可诱导BGC-823细胞凋亡,实现G0/G1期细胞周期阻滞。3.诱导凋亡的机制可能与下调Ezrin和上调ICAM-1的表达有关。
