人参皂苷Rg1对局灶性脑缺血再灌注自噬及其相关基因Beclin-1、LC3、mTOR表达的影响

摘要
目的探讨在大鼠局灶性脑缺血再灌注(cerebral ischemia reperfusion,CIR)后,人参皂苷Rg1对哺乳动物雷帕霉素(mammalian rapamycin,m TOR)、微管相关蛋白1轻链3(microtubule-associated protein 1 light chain 3,LC3)、bcl-2同源结构域蛋白(bcl-2 homeodomain protein,Beclin-1)在各组别脑组织顶叶中的表达规律,在此基础上探讨人参皂苷Rg1对局灶性脑缺血再灌注自噬损伤的调控。方法将健康的雄性大鼠随机分为四组,分别为假手术组(sham组)、模型组(IR组)、人参皂苷Rg1给药组(Rg1组)和人参皂苷Rg1加自噬抑制剂3-甲基腺嘌呤(3-methyladenine,3-MA)给药组(Rg1+3-MA组)。大鼠大脑局灶性缺血再灌注模型采用大脑中动脉栓塞(Middle cerebral artery occlusion,MCAO)2h再灌注24h的方法制备。通过观察各组间的神经功能缺损评分、脑组织含水量测定和2,3,5-三苯基氯化四氮唑(2,3,5-triphenyltetrazolium chloride,TTC)染色结果鉴定模型是否成功。运用透射电镜观察各组别间大鼠脑组织神经元中的自噬情况;采用免疫荧光和免疫印迹定性定量检测各组间大鼠大脑顶叶m TOR、LC3、Beclin-1的表达。结果神经功能缺损评分结果示,sham组未见神经功能缺损症状;其余各组可见明显的神经功能缺损症状。脑组织含水量测定结果示,与sham组相比,其余各组的脑组织含水量明显增加。TTC染色结果示,sham组无梗死,其余各组可见明显苍白色梗死灶。与Rg1组相比,IR组和Rg1+3-MA组苍白色梗死灶体积明显增大。透射电镜结果示,sham组未见损伤的神经元形态;其余各组可见明显的神经元自噬。免疫荧光和免疫印迹结果示,IR组与Rg1+3-MA组无明显的差异。与sham组相比,其余各组间Beclin-1、LC3表达增加,m TOR表达降低,且在Rg1组中Beclin-1、LC3表达最高,m TOR表达最低。结论人参皂苷Rg1可能通过调节大脑顶叶Beclin-1、LC3和m TOR的表达水平,使脑组织细胞自噬水平上调,进而对大鼠局灶性脑缺血再灌注神经元起到保护作用。
