血清和糖皮质激素诱导的蛋白激酶1对大鼠肾动脉平滑肌细胞增殖的作用及其分子学机制

摘要
目的:探讨血清和糖皮质激素诱导的蛋白激酶1(SGK1)肾动脉平滑肌细胞(VSMCs)增殖中的作用,探讨SGK1在高血压肾损害中的可能作用机制。 方法:组织块贴壁法原代培养的大鼠平滑肌细胞(VSMCS),将带有SGK1显性激活型突变体质粒(pIRES2-EGFP-S422D SGK1 mutant SD),采用细胞计数,细胞流式技术,MTT比色法分析,来观察转染后平滑肌细胞增值情况,应用Western Blot方法检测SGK1对细胞周期调控蛋白p27kip1、p21cip1,增殖细胞核抗原(PCNA)和磷酸化糖原合成酶激酶3(P-GSK3)的作用。 结果:SGK1能刺激平滑肌细胞增值,使滞于G0/G1期平滑肌细胞减少,周期调控蛋白p27kip1、p21cip1表达下降,PCNA,P-GSK3表达升高。 结论:SGK1介导平滑肌细胞增值,SGK1可能通过介导平滑肌细胞增殖参与了早期高血压性肾损害。
