人参花酸性多糖GFLA-1的纯化及其分析

本实验室的前期工作对人参花总多糖进行了提取分离,并对其部分级分进行了结构鉴定及活性研究。在此基础上,本文对人参花酸性糖级分GFLA-1进行制备,将其进一步分离纯化,同时研究了各级分的理化性质、单糖组成及结构特征。 干燥的人参花经过热水煮提、乙醇醇沉精制得到人参花水溶性粗多糖GFL(79.1 g)。高效液相色谱法分析GFL的单糖组成,其单糖摩尔比为Man:Rha:GlcA:GalA:Glc:Gal:Xyl:Ara=3.1:2.2:0.9:16.0:8.2:52.3:1.5:10.8。GFL经过DEAE-Cellulose柱层析分级,得到一个中性糖级分GFLN和四个酸性糖级分GFLA-1、GFLA-2、GFLA-3、GFLA-4,并将这五个级分与实验室之前研究得到的样品进行了含量、单糖组成以及分子量的比较分析。因之前的工作未对GFLA-1深入研究,故本论文将GFLA-1经Sepharose CL-6B柱层析作进一步分级,得到分子量不同的两个级分GFLA-1-A和GFLA-1-B。 GFLA-1-A经过高效凝胶渗透色谱和Sepharose CL-6B分析显示呈单一对称的峰,分子量约为9.1×104 Da。高效液相色谱法分析其单糖组成成分,结果显示GFLA-1-A主要由半乳糖和阿拉伯糖组成,含量分别为58.3%和34.4%,比例接近2:1,且两者之和占总含量的90%。另外,还含有少量的鼠李糖(4.4%)、半乳糖醛酸(2.3%)和甘露糖(0.6%)。成分中阿拉伯糖与半乳糖构成阿拉伯半乳聚糖结构域,同时因为有少量鼠李糖和半乳糖醛酸的存在并且比例接近,推测GFLA-1-A是一种AG型中性多糖,并含有RG型侧链。 GFLA-1-B经高效凝胶渗透色谱及Sepharose CL-6B分析同样呈现出单一窄峰,总糖和糖醛酸峰均表现出较好的对称性,分子量分布较集中,约为4.7×103 Da。GFLA-1-B的单糖组成比较复杂,所含单糖种类比较繁多。其最主要的单糖为半乳糖,含量高达35.6%。再者,半乳糖醛酸也是主要的单糖,含量占总含量的27.3%。除以上两者单糖外,阿拉伯糖占总糖的17.9%。这三种单糖含量达到GFLA-1-B总糖的80%,另外还有鼠李糖(5.8%)、葡萄糖醛酸(4.9%)、葡萄糖(4.5%)和甘露糖(3.0%)。其中,半乳糖:阿拉伯糖约为2:1,推测其中同时含有阿拉伯半乳聚糖和聚半乳糖结构域。又由于少量鼠李糖和半乳糖醛酸的存在,推测其中应含有少量RG型侧链。
