半夏白术天麻方抗抑郁作用与大鼠脑海马BDNF表达的实验研究

摘要
目的:通过分析抑郁模型大鼠行为学及海马组织内脑源性神经营养因子的改变,探讨中药方剂抗抑郁作用及机制。方法:60只SD大鼠,随机分为空白对照组、模型对照组、中药方剂干预组(低、中、高剂量组)、西药组,每组10只,雌雄对半。除空白对照组外均采用慢性不可预见性应激和孤养法建立抑郁症动物模型,空白对照组每日灌服生理盐水一次,模型对照组造模过程中每日灌服生理盐水一次,中药方剂干预组造模过程中每日分别灌服低、中、高剂量的中药方剂持续4周,西药干预组同样每日灌服氟西汀。观察大鼠体重的变化,记录旷场试验和液体消耗实验,最后取海马组织,采用荧光定量PCR法,测定各组大鼠脑内海马区BDNF因子的表达。结果:4周后模型对照组较空白对照组,体重降低,糖水消耗量,旷场试验水平及垂直运动得分明显减低(P<0.01)。各给药组较空白对照组上述评分接近,差异无统计学意义(P>0.01)。与空白对照组相比,模型对照组海马组织经荧光定量PCR法测定BDNF因子的表达水平与之相比出现差异(P<0.01),各给药组较空白对照组BDNF因子水平接近(P>0.01),中药(中、高剂量)组、西药组干预下大鼠海马区BDNF因子的表达水平差别不具有统计学意义。结论:半夏白术天麻方在一定剂量使用后与西药均能够有效干预抑郁症动物模型,其机制可能与脑内BDNF因子表达增多,从而发挥外在行为学改变。
