赤萆颗粒制备工艺和质量标准研究

摘要
目的:急性痛风性关节炎(GA)是痛风的首发症状,会导致关节畸形及功能障碍;重者会引起痛风性肾病、冠心病甚至死亡。中药无论是在抗炎镇痛、调整机体免疫功能,改善症状与体征;还是在促进尿酸排泄、改善血液流变、保护关节等方面,都发挥着极其重要的作用。本课题选取临床经验方,在中医药理论指导和最新药理研究进展并重的基础上,采用先进的筛选及优化设计理念,优选出最佳提取和分离工艺,为将其开发成中药成方制剂奠定药学研究基础。 方法:(1)制剂工艺研究:应用单因素考察和正交实验设计,以芍药苷、薯蓣皂苷元、牛蒡苷、齐墩果酸为指标成分,分别考察并确立赤芍、萆薢、络石藤和牛膝的提取工艺条件;用天然絮凝剂壳聚糖,以总皂苷元和总多糖含量为指标,运用星点设计结合效应面法确定牛膝水煎液的最佳絮凝沉淀纯化工艺,并对牛膝壳聚糖絮凝沉淀的动力学特征进行了初步研究。将上述干浸膏合并,粉碎,过筛,加适量乳糖,混匀,用5%丙烯酸树脂乙醇溶液作黏合剂,湿法制粒,干燥,整粒分装后,即得成品。并初步考察了药粉以及成品的吸湿特性。(2)质量标准研究:采用HPLC法对芍药苷进行定量测定;采用TLC法分别对赤芍、萆薢、络石藤和牛膝进行定性鉴别。 结论:本研究通过提取方中各药的有效部分,研究开发成颗粒剂,并进行制剂质量标准研究,为临床治疗痛风性关节炎提供疗效确切、质量稳定可控的中成药,奠定了药学实验基础。
