AAPⅠ-a黑木耳多糖的分离纯化及其抗衰老功能的研究

黑木耳(Auriculaia Auricula)隶属于真菌门担子菌纲,是我国重要的药、食兼用菌,其主要活性成分——黑木耳多糖具有调节人体免疫系统、降血脂、降血糖、抗肿瘤、抗凝血等功效。本文以黑木耳子实体为原料,对其提取工艺、分离纯化、理化性质及抗衰老活性进行了研究。 采用超声波辅助提取黑木耳多糖,响应面分析法优化提取工艺,得到黑木耳多糖的最佳提取条件为:超声功率520W,超声时间13min,液料比132:1,水浴浸提时间3.1h,多糖得率为16.59%。 精多糖经过CTAB沉淀法分级、DEAE Sephadex A-25离子交换及Sephadex G-200排阻层析,得到三个主要组分,分别命名为AAPⅠ-a、AAPⅡ-a及AAPⅢ-a。分别采用薄层层析法、冻融分析法和高效液相色谱(HPLC)对黑木耳多糖的三个主要组分进行纯度鉴定,结果表明AAPⅠ-a及AAPⅡ-a为单一组分,而AAPⅢ不是单一组分,AAPⅠ-a、AAPⅡ-a及AAPⅢ-a的纯度分别为91.12%、90.03%及89.98%。 经GPC测定,AAPⅠ-a的(M|-)w为2.06×10~5,(M|-)_n为7.41×10~4;AAPⅡ-a的(M|-)w为1.86×10~4,(M|-)_n为1.16×10~4;AAPⅢ-a的(M|-)w为1.24×10~5,(M|-)_n为2.61×10~4。经红外光谱分析,AAPⅡ-a为β-D-葡萄吡喃糖, AAPⅠ-a及AAPⅢ-a同时具有α-构型和β-构型,是既有葡萄糖又有半乳糖的D-吡喃糖。经GC/MS测定,AAPⅠ-a由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖及半乳糖组成,各单糖之间比例为0.2:2.6:0.4:3.6:1.0:0.4。 AAPⅠ-a黑木耳多糖对小鼠体重影响不显著,但可使胸腺指数及脾指数升高,显著增高超氧化物歧化酶(SOD)的活力及谷胱甘肽过氧化物酶(GSH-PX)的活力,降低过氧化产物丙二醛(MDA)的含量,揭示了AAPⅠ-a黑木耳多糖具有明显的抗衰老功效。
