双歧杆菌BB12治疗炎症性肠病及其作用机制研究

炎症性肠病(inflammatory bowel disease, IBD)包括克罗恩氏病(Crohn’s Disease, CD)和溃疡性结肠炎(Ulcerative Colitis, UC),其病因目前仍不清楚,但肠道菌群在IBD发病中的作用日益引人注目。有研究提示IBD患者存在肠道菌群失调,益生菌数量明显减少,致病菌、条件致病菌明显增多。进一步研究发现,肠道内菌群失调与IBD患者肠道免疫功能紊乱密切相关,纠正肠道菌群失调可能会缓解IBD病情。双歧杆菌是人体肠道内主要的生理性益生菌,对维持肠道微生态平衡及人体健康起着重要作用。 为深入了解益生菌双歧杆菌BB12参与IBD肠道炎症反应的机制,本研究从三个方面进行探讨: 1.以致病性肠炎沙门氏菌和炎症细胞因子分别刺激体外培养的肠上皮细胞系Caco-2细胞,模拟肠道炎症反应,探讨肠道炎症反应中双歧杆菌BB12对肠上皮细胞功能的调节及其抗炎机制。然后通过体内实验,进一步了解双歧杆菌BB12抗炎作用的有效性。 2.建立了与人类克罗恩氏病十分相似的TNBS诱发的大鼠结肠炎模型,探讨双歧杆菌BB12对实验性结肠炎的防治作用和可能的机制。 3.体外共同培养双歧杆菌BB12与克罗恩氏病患者结肠粘膜活检组织,以探讨双歧杆菌BB12对克罗恩病的治疗效果及可能的机制。
