唑来膦酸对鼠骨质疏松性骨折愈合及TGF-β表达影响的实验研究

摘要
目的研究唑来膦酸对去势大鼠骨质疏松性骨折愈合过程中转化生长因子(TGF-β)表达的影响,从而获得唑来膦酸、TGF-β和骨折愈合之间的联系,为今后骨质疏松骨折的治疗和促进骨质疏松骨折愈合提供依据。 方法采用60只6月龄健康雌性SD大鼠,将其随机分为对照组、实验组,每组25只,假手术组、空白组,每组5只。实验组和对照组采取卵巢去势法建立骨质疏松模型并确认成功后,在麻醉下制作左股骨中断骨折模型,实验组术后即刻皮下注射给予浓度为100μg/kg的唑来膦酸;对照组分别给予同体积0.9%氯化钠注射液同一部位皮下注射。各组分别于术后不同时间段取骨痂,行苏木素-伊红(HE)染色、测骨密度、免疫组化及X线片观察骨折愈合情况。分析唑来膦酸与大鼠骨质疏松骨折愈合之间的相关性。 结果①放射学检查:唑来膦酸组比同时期对照组骨痂生成量多,愈合时间提前。②骨密度:骨折局部骨密度明显高于对照组,差异具有显著统计学意义(P<0.05)。③TGF-β表达:于4周左右达到高峰,唑来膦酸组比对照组TGF-β表达更明显(P<0.05)。 结论唑来膦酸能加快骨痂形成,促进成骨细胞TGF-β的表达,TGF-β是唑来膦酸调节骨代谢的机制之一,可促进骨质疏松骨折性的愈合。
