BMP7调节和厚朴酚抗结肠癌细胞增殖作用与p53的关系研究

摘要
和厚朴酚(Honokiol)作为抗肿瘤的中药活性成分之一,近些年来越来越受到研究学者们的青睐。但其确切的抗癌机制还没有完全被阐释。在本研究中,我们利用了一些基本的分子生物学技术证实了和厚朴酚可以有效地抑制结肠癌细胞的增殖并诱导其凋亡。研究发现,和厚朴酚发挥抗肿瘤作用可能与BMP7的蛋白水平和cDNA水平上调有关,并在其他类型的结肠癌细胞中(SW620与LoVo)进行了验证,其结果与和厚朴酚在HCT116细胞中的结果一致。利用重组腺病毒技术组装成BMP7外源性过表达腺病毒(AdBMP7)。过表达BMP7能进一步增强和厚朴酚抗肿瘤细胞增殖的作用;其抗体则部分减弱了和厚朴酚的作用。通过进一步筛选得知,细胞中p53的表达也与和厚朴酚的浓度及其作用时间呈正相关关系,并在SW620与LoVo细胞中进行了验证,其结果与和厚朴酚在HCT116细胞中一致。P53外源性过表达腺病毒(Adp53)能够增强和厚朴酚的作用,其抑制剂则部分减弱了和厚朴酚的作用。机制分析:和厚朴酚对HCT116细胞中的Smad1/5/8和磷酸化的Smad1/5/8并没有明显的影响,说明和厚朴酚可能不是通过经典的BMPs/Smad信号通路发挥作用的。实验结果提示,和厚朴酚可能是通过BMP7上调p53的表达来调节其对结肠癌细胞的作用。
