圣脑康丸对大鼠脑缺血—再灌注损伤的保护及降压作用研究

摘要
脑缺血/再灌注损伤及高血压均是常见的心脑血管疾病,中药复方从多靶点、多向调节出发对心脑血管疾病起保护作用。圣脑康丸(SNKP)由丹参、川芎等15味中药组成,具有活血化瘀、凉血降压等功效,临床用于急、慢性心脑血管病的治疗,目前尚未有SNKP对脑缺血/再灌注损伤保护及降压的实验研究报道。 本实验采用HPLC法同时测定了SNKP中丹参素、原儿茶醛、芍药苷、葛根素、阿魏酸、丹参酮ⅡA6种有效成分的含量,甲醇-0.25%冰醋酸水溶液为流动相梯度洗脱,检测波长为280nm,方法简便、准确、灵敏。 采用线栓法制作大鼠局灶性脑缺血/再灌注损伤模型和小鼠常压耐缺氧实验,研究了SNKP对脑缺血缺氧的保护作用。通过大鼠神经功能评分、TIC染色、H&E染色法等方法进行综合评价;同时测定大鼠血清或组织中MDA、SOD、GSH-Px、iNOS、Caspase-3等相关指标活性,观察小鼠常压耐缺氧时间,结果表明SNKP能明显改善神经功能障碍,减少大脑梗死体积,增加存活的神经元数量,降低MDA含量,增强SOD、GSH-Px活性,降低iNOS及TNOS活性,减少Caspase-3活性细胞数目。说明SNKP对脑缺血/再灌注损伤有保护作用,其保护机制可能与其抗氧化、抗凋亡与降低iNOS和TNOS活性等有关。SNKP能有效提高常压下及异丙肾上腺素联合给药的耐缺氧时间,具有明显的耐缺氧作用。 利用自发性高血压大鼠研究了SNKP的降压作用。采用无创血压测量分析系统测量血压及心率,采用全自动生化分析仪测定血脂、肾功能等相关的生化指标,并测量了各脏器指数、左心室指数等,结果表明SNKP能有效平稳的降低血压,调节心率,降低左心室指数,改善肾功、血脂等生化指标,恢复对主要器官(心、脑、肾等)的损伤。 因此,SNKP对大鼠脑缺血/再灌注损伤有保护作用,对自发性高血压大鼠有降压作用,并能有效延长小鼠的常压耐缺氧时间,为心脑血管疾病的治疗提供了理论基础。
