木犀草素对活化T细胞NF-κB及IκBa的调节作用

目的 本研究研究了痹肿消汤有效成分之一木犀草素体外对T淋巴细胞增殖以及NF-κB信号通路的影响,探讨痹肿消汤有效成分木犀草素治疗类风湿关节炎的抗炎、抑制免疫的作用机制。 方法 佛波醇酯(PMA)+离子霉素(ionomycin,Ion)刺激小鼠脾淋巴细胞来源的T细胞,以不同终浓度的木犀草素与T细胞共培养,实验分为T淋巴细胞组、T淋巴细胞+PMA+Ion组、T淋巴细胞+PMA+Ion+10μM木犀草素组、T淋巴细胞+PMA+Ion+20μM木犀草素组、T淋巴细+PMA+Ion+30μM木犀草素组共五组,用MTT法测定细胞增殖;采用Western blot方法检测药物对PMA加Ion刺激作用下的T细胞IκB-α磷酸化和P65入核的影响。 结果 (1) MTT法结果显示PMA加Ion刺激48h后明显促进T淋巴细胞的增殖,木犀草素(10-30μM)能抑制小鼠T淋巴细胞活化与增殖(P<0.01)。 (2) Western blot结果显示p65在PMA+Ion刺激T细胞30min时入核最多,木犀草素(10-30μM)能抑制活化的T细胞细胞核内p65表达(P<0.01),且呈一定的剂量依赖性;PMA+Ion刺激T细胞12 h时,IκB磷酸化的表达最强,木犀草素(10-30μM)能抑制活化的T细胞IκB的磷酸化(P<0.01),且呈一定的剂量依赖性。 结论: (1)木犀草素能抑制小鼠T细胞的活化与增殖,提示木犀草素具有免疫抑制作用。 (2)木犀草素能抑制T细胞NF-κB信号通路,提示木犀草素可能通过NF-κB信号通路来抑制T细胞的功能,从而达到抑制免疫的作用。
