泽泻对奶牛脂肪变性肝细胞CYP450的影响

摘要
脂肪肝是奶牛常见疾病,一般围产期高产奶牛多发。脂肪肝会导致奶牛产奶量下降,并发多种其他疾病,给养牛业带来巨大的经济损失。近些年来,奶牛的脂肪肝发病率成逐年上升趋势,但是对于脂肪肝的发生发展机制尚不明确。细胞色素酶P450(CYP450)是肝脏微粒体氧化酶系的主要代谢酶,在肝脏的代谢中起着重要的作用。 本研究通过胶原酶两步灌流法分离培养奶牛的原代肝细胞,用DL-乙硫氨酸诱导肝细胞脂肪变性,测定谷丙转氨酶(ALT)、谷草转氨酶(AST)、超氧化物歧化酶(SOD)的活性,以及甘油三酯(TG)、总胆固醇(TC)、游离脂肪酸(FFA)、丙二醛(MDA)的含量。油红O染色肝细胞,观察肝细胞形态变化,及细胞内脂滴变化。根据以上生化指标和肝细胞形态变化特征,制备肝细胞脂肪变性的模型。用不同浓度的泽泻处理变性肝细胞24h-48h,检测细胞上清液中ALT、AST、SOD的活性和细胞裂解液中TG、TC、FFA、MDA的含量,双抗体夹心ELISA法检测TNF-α的含量,油红O染色观察肝细胞形态,Western blot法分析肝细胞CYP450的表达。研究CYP450在奶牛脂肪肝形成中的作用,探讨奶牛脂肪肝的形成机制,为奶牛脂肪肝的治疗提供基础。 结果表明,胶原酶灌流法分离到的肝细胞形态完整,生长良好,可用于脂质代谢的研究。DL-乙硫氨酸诱导肝细胞后,ALT、AST活性增强,SOD活性减弱,TG、TC、FFA、MDA含量升高,脂滴明显增多,变大。诱导脂肪变性的最佳条件是2mmol/LDL-乙硫氨酸作用肝细胞24h。泽泻处理脂肪变性肝细胞后,ALT、AST活性减弱,SOD活性增强,TG、TC、FFA、MDA和TNF-α的含量降低,脂滴明显减少,体积变小。Western blot结果显示,添加不同浓度的泽泻以后,模型组肝细胞CYP450的表达逐渐减弱。 2mmol/L DL-乙硫氨酸作用肝细胞24h可成功制备脂肪变性肝细胞模型,10μg/mL泽泻作用48h,对脂肪变性肝细胞具有较好的保护作用。
