低温厌氧生境中产甲烷古菌的分离、鉴定及菌群结构分析

摘要
低温厌氧消化技术在实现废弃物资源循环利用方面发挥着很重要的作用。本实验对在厌氧消化过程中起关键作用的产甲烷古菌进行了研究,同时采用PCR-DGGE技术研究了低温厌氧消化系统中微生物群落的结构变化。 采用Hungate厌氧操作技术,从西藏林芝和堆龙德庆地区厌氧消化系统中分离出三株兼性嗜冷产甲烷古菌,即LZ-6、DL-4和DL-5。通过16S rDNA分析,菌株LZ-6属于甲烷粒菌属(Methanocorpusculum)的小甲烷粒菌(序列相似性99%)。对LZ-6的生物学特性进行了研究。菌体呈小的不规则球形,最适生长温度25℃,利用H2/CO2,微利用甲酸,不利用乙酸、甲醇。 本文采集了厌氧消化系统中不同月份的样品,提取基因组总DNA,分别扩增细菌和古菌的16S rDNA V3区。DGGE图谱的初步分析结果表明,不同月份的样品的DGGE带谱在条带数量、位置及条带亮度上存在较大差异。Quantity One软件定量分析表明,厌氧消化系统的细菌、古菌群落组成主要分为四种类型,即秋初期、秋末期、冬末期和春初期。典型条带测序结果显示,存在于每个样品中的优势细菌类群为Uncultured bacterium clone FLSED43和Uncultured rumen bacterium clone YRC75。产甲烷古菌菌群主要类群为甲烷粒菌属(Methanocorpusculum)。
