VEGF、HIF-1α在大鼠哮喘模型中的表达及柴朴汤的干预研究

目的:观察缺氧诱导因子、血管内皮生长因子在哮喘大鼠中的表达水平,分析其相互关系及其与气道重塑的关系,探讨其在哮喘气道重塑中的作用。同时研究柴朴汤治疗支气管哮喘的可能作用机制。
方法:32只SD大鼠随机分成哮喘模型组(A)、柴朴汤干预组(B)、地塞米松干预组(C)、正常对照组(D)四组。A、B、C组以腹腔注射卵蛋白(OVA)致敏及反复雾化吸入OVA1%激发建立大鼠哮喘模型,B、C两组激发前30分钟分别用地塞米松1mg/kg和柴朴汤7ml/kg灌胃进行干预,最后一次激发12 h内处死大鼠。常规病理切片观察各组大鼠肺内炎症情况、测量各组大鼠气管总管壁厚度、气管内管壁厚度及气道平滑肌厚度。RT-PCR观察大鼠肺部HIF-1α、VEGFmRNA的表达,免疫组化检测气道壁HIF-1α、VEGF蛋白的表达情况。比较各组大鼠HIF-1α和VEGF的表达差异,分析HIF-1α与VEGF的关系,HIF-1α、VEGF与气道壁厚度的关系。
结果:1.大鼠肺组织常规病理切片示:A组大鼠气道支气管及血管周围可见大量炎症细胞浸润,以嗜酸性粒细胞、淋巴细胞为主,部分气道上皮脱落,粘膜下细胞外基质沉积,B、C组上述改变较轻,D组无上述改变。2.图像分析显示A组总气管壁厚度标化值(Wat/Pbm)、气管内壁厚度标化值(Wai/ Pbm)及气道平滑肌层厚度标化值(Wam/Pbm)较D组大,B、C组介于两者之间。气管壁各层厚度标化值分别为Wat/Pbm(μm2/μm)(A组68.66±5.22、B组50.94±3.86、C组42.82±3.84、D组34.80±3.48),Wai/Pbm(μm2/μm)(A组.38.21±4.08、B组26.32±3.22、C组21.64±3.12、D组16.51±1.24), Wam/Pbm(μm2/μm)(A组8.04±0.90、B组4.64±0.86、C组3.22±0.62、D组2.96±0.44)。3.免疫组化蛋白半定量分析,A组HIF-1α、VEGF蛋白的积分光密度值(IOD)较D组明显增高,B、C组IOD值介于两者之间,它们分别为(82.26±6.06、71.12±5.20、65.58±4.88、55.42±4.66), (96.31±5.64、82.62±5.44、76.86±5.22、68.74±4.34)。4.RT-PCR半定量分析A组HIF-1α、VEGFmRNA相对灰度值较D组明显增高,B、C组介于两者之间。它们分别为(0.60±0.15、0.46±0.10、0.39±0.10、0.28±0.08),(0.70±0.14、0.59±0.12、0.51±0.12、0.42±0.10)。5.相关性分析显示,哮喘模型大鼠气道VEGF与HIF-1α蛋白的积分光密度值呈正相关,VEGFmRNA与HIF-1αmRNA相对灰度值呈正相关。
结论:1)哮喘大鼠HIF-1α、VEGF表达水平增高,HIF-1α、VEGF可能参与了哮喘大鼠的气道重塑。
2)早期使用柴朴汤治疗可以下调HIF-1α、VEGF的表达,减轻气道重塑。
