哈药包尔胡特提取物对大鼠酒精性肝病的作用

摘要
目的:对包尔胡特的化学成分进行初步研究,并筛选包尔胡特的有效部位;通过建立大鼠酒精性肝病动物模型探讨包尔胡特提取物(RSE)对酒精性肝病(ALD)大鼠的作用。方法:采用植物化学成分系统预试和专项预试相结合的方法进行化学成分初步研究;采用小鼠急性酒精性肝损伤模型筛选包尔胡特有效部位,并建立大鼠酒精性肝病动物模型,阳性对照组以100mg.kg-1剂量给予益肝灵,包尔胡特提取物小、中、大剂量组分别按照0.9,1.8,3.6g.kg-1剂量给药。给药4周后,测定血清和肝组织有关生化指标,并观察肝脏组织病理学改变。结果:包尔胡特在系统预试试验中糖类、鞣质、皂苷、醌类、香豆素内酯类、有机酸、酚类及油脂为阳性,在专项预试试验中蒽醌类为阳性。包尔胡特的95%乙醇部位对酒精性肝损伤有明显干预和治疗作用。RSE对大鼠酒精性肝病的作用:与正常对照组比较,酒精性肝病大鼠血清中ALT、AST、TG、TC、LDL-C含量显著性升高(P<0.01),肝组织中T-SOD活性显著降低(P<0.01),MDA、NO含量明显升高(P<0.01),肝组织明显脂肪变性。给予RSE后,与模型组比较,包尔胡特组血清中ALT、AST活性和TG、TC、LDL-C含量较模型组显著降低(P<0.01)。肝组织中MDA、NO含量较模型组明显降低(P<0.05),大剂量组T-SOD活性较模型组显著升高(P<0.05),肝组织病理改变明显好转。结论:包尔胡特中含有糖类、鞣质、皂苷、醌类、香豆素内酯类、有机酸、酚类及油脂等成分。包尔胡特提取物通过增强机体清除自由基、抗脂质过氧化作用的能力改善脂质过氧化反应对肝脏的损伤和保护肝细胞的结构和功能,发挥对酒精性肝病的治疗作用。
