摘要
利用基质辅助激光解吸附飞行时间质谱(MALDI-TOF MS)和反相高效液相色谱(RP-HPLC)分析技术探索和完善快速、准确的分析植物多酚结构与含量的方法,筛选杨梅、茶叶和柚子中能用于药物中间体和保健品开发的植物多酚种类,得到如下研究结果: 1.利用RP-HPLC快速、准确的测定柚子果皮中4种具有良好生理学和药理学活性的多酚化合物含量。结果表明,柚子果皮总酚含量为23.45~26.97mg/g(儿茶素当量),4种化合物含量分别为4.02~4.84 mg/g(柚皮苷),2.41~2.90 mg/g(对羟基肉桂酸),0.16~0.31 mg/g(橙皮素),0.010~0.012mg/g(桔皮素);当体积浓度为60%~70%时,甲醇水溶液与乙醇水溶液对柚子果皮总酚的提取效果没有显著差异,但是前者提取的黄酮类化合物含量明显高于后者。 2.首次通过MALDI-TOF MS揭示了茶叶缩合单宁的基本结构,并测定了本山、黄旦和铁观音三个品种茶树成熟叶片的总酚及可溶缩合单宁含量。结果显示,三种茶叶均具有较高的总酚含量,大约为200mg/g;三种茶叶的缩合单宁是以(表)儿茶素(EC/C)和(表)桔儿茶素(EGC/GC)为基本结构单元的均聚物和杂聚物,且大部分聚合物的结构单元之间存在A型和B型两种连接方式,但其中本山和黄旦缩合单宁的最高聚合度要大于铁观音。 3.首次利用MALDI-TOF MS详细研究了杨梅不同器官中缩合单宁的类型、聚合度及聚合物的分布情况,并对叶、枝和树皮的总酚含量与可溶缩合单宁含量进行了测定。结果表明:树皮的总酚含量最高(435.62±0.66 mg/g),叶次之(346.87±0.82 mg/g),枝最低(185.29±20.21 mg/g);树叶的可溶缩合单宁含量最高(256.71±13.76 mg/g),皮次之(179.28±13.16 mg/g),枝最低(80.07±18.45 mg/g);3组分的缩合单宁都以(表)棓儿茶素-3-O-棓酸酯(EGCG/GCG)为基本结构单元的原翠雀定,且大部分聚合物的结构单元之间存在A型和B型两种连接方式,但其中叶和枝缩合单宁的聚合度要大于树皮。
