七氟烷通过抑制Grp78的表达减轻大鼠肝缺血再灌注损伤

摘要
目的研究七氟烷预处理在肝缺血再灌注的作用及其潜在机制是否与Grp78的表达相关。方法选取雄性大鼠24只,平均重量240±15克,饲养一周后,随机分为三组:对照假手术组(Sham组),模型组(I/R组),七氟烷组(SEV组)。对照组麻醉后仅进行剖腹及二次剖腹,不进行肝脏缺血再灌注处理,I/R组和SEV组建立大鼠肝脏缺血再灌注模型。SEV组使用七氟烷预先处理,完成手术后,采集三组大鼠腔静脉血液样本,离心获得上层液。迅速采集肝脏样本,使用0.9%盐水与100g肝组织制成10%匀浆。通过酶联免疫法测定血清中TNF-α、IL-1β、IL-10和IL-6的浓度。使用检测盒测定肝均质中丙二醛(MDA),超氧化物异变酶(SOD)和一氧化氮(NO)的含量。使用HE染色、末端标记法和免疫组化等方式观察细胞损伤程度、细胞凋亡及Grp78的表达。培养小鼠胚胎肝细胞BNLCL.2,在细胞对数生长阶段收集细胞,根据不同方式分为对照组(Control组)、模型组(I/R组)和七氟烷组(SEV组)。使用糖氧剥夺方式建立了肝细胞缺血再灌注模型,SEV组预先使用七氟烷处理,收集细胞并进行细胞转染。转染后,对照组细胞分为siGrp78细胞组和siNC细胞组。I/R组的细胞,并分别命名为I/R+siNC组和I/R+siGrp78组。同时,将SEV组细胞分为I/R+siNC+SEV组和I/R+siGrp78+SEV组。使用流式细胞术观察细胞凋亡,定量实时PCR(qRT-PCR)测定Grp78 mRNA含量和采用Western Blot分析PERK,eIF2α,p-c-JNK/JNK蛋白表达。结果与I/R组比较,SEV组大鼠中肝损伤程度、肝细胞凋亡、葡萄糖调节蛋白78(Grp78)的表达明显降低。TNF-α、IL-1β和IL-6浓度也显著降低(P<0.01)。MDA和NO浓度显著较低(P<0.01),IL-10和SOD浓度明显增高(P<0.01)。与I/R组细胞比较,SEV组细胞凋亡率、Grp78、PERK、eIF2α和p-c-JNK/JNK的表达显著下降(P<0.01),仍然高于对照组细胞。siGrp78组BNLCL.2细胞中siGrp78 mRNA和蛋白表达显著低于siNC组(P<0.01)。对于siNC组的细胞,I/R+siNC组凋亡率高于siNC组(P<0.01)。I/R+siNC+SEV组的细胞凋亡率则低于I/R+siNC组(P<0.01)。与siGrp78组相比,I/R+siGrp78组细胞凋亡率明显增加(P<0.01)。I/R+siGrp78组和I/R+siGrp78+SEV组之间没有发现凋亡率的明显差异。蛋白质印迹分析显示与siNC组相比,在I/R+siNC组中发现PERK,eIF2α和p-c-JNK/JNK蛋白表达显著升高(P<0.01)。而I/R+siNC+SEV组的表达均低于I/R+siNC组(P<0.01)。同时观察到I/R+siGrp78组中PERK,eIF2α和p-c-JNK/JNK蛋白的相对表达组比siGrp78组高很多(P<0.01)。观察I/R+siGrp78+SEV组与I/R+siGrp78组相比蛋白质相对表达的变化不明显。七氟烷通过抑制Grp78(P<0.01)的表达显著降低细胞凋亡和PERK、eIF2α、p-c-JNK/JNK的表达。结论七氟烷通过降低Grp78的表达来抑制肝缺血再灌注损伤。
