电针对大鼠急性脊髓损伤后胶质纤维酸性蛋白表达影响的实验研究

摘要
目的:通过观察电针治疗脊髓损伤前后脊髓组织中GFAP表达的变化情况来阐明电针治疗脊髓损伤的机理。 方法:Wistar大鼠60只,随机分为:对照组(A组)20只,模型组(B组)20只和电针治疗组(C组)20只。采用改良的Allen's法建立SCI模型。治疗采用KWD-808Ⅱ型脉冲电针仪,选择连续密波,频率100Hz,电压9V,在T9~T10和T10~T11棘突间隙旁开距中线3-4mm处取穴,操作时电针仪的二组导线分别上下连接针柄,正极在上,负极在下,然后打开电流输出旋钮,电流输出强度以背部肌肉出现轻微抽动为度。每日治疗2次,每次15分钟。最后分别于不同的时间点:术后3d,7d,14d和21d取损伤部位的脊髓组织,采用免疫组化、蛋白印迹(Western Blot)等方法和光镜技术来检测脊髓组织中的GFAP表达的变化,采用BBB评分方法对各组实验大鼠后肢运动功能进行评估。 结果: 1.BBB评分结果显示:脊髓损伤后3d,大鼠BBB评分C组优于B组,统计学有显著差异(P<0.05)1w开始,C组大鼠明显优于B组,统计学有非常显著差异(P<0.01),但两组评分均低于A组(P<0.01)。 2.免疫组化染色结果显示:脊髓损伤后经电针治疗后3d,7d,大鼠C组星形胶质细胞数明显低于B组,统计学有非常显著差异(P<0.01),14d后C组星形胶质细胞数仍低于B组,统计学有显著差异(P<0.05),但21d后C组星形胶质细胞数与B组相近,统计学无显著差异(P>0.05). 3.Western Blot结果显示:各组脊髓均有GFAP蛋白表达。B组GFAP蛋白表达呈递增趋势。2w后表达最强,以后逐渐降低,B组表达明显高于A,C组。 结论: 1.电针能减少星形胶质细胞增生,从而创造了有利于脊髓损伤后功能恢复的微环境。因此证明是治疗脊髓损伤的有效手段。 2.电针能够促进脊髓损伤后功能的恢复,其机理可能是通过抑制GFAP的表达而起作用。 3.脊髓损伤后2周内可能为修复损伤的一个时间窗口,提示越早治疗效果越好。
