蛭龙通络片对大鼠脑缺血再灌注损伤的保护作用

蛭龙通络片以0.405、0.81、1.62g/kg剂量给大鼠连续灌胃7天,末次给药后1h,采用Zea longa线栓法制作大鼠大脑中动脉缺血再灌注模型,缺血2h,再灌注24h。再灌注24h后进行Zea5分制神经功能学评分;测量大鼠脑组织含水量、脑指数;检测全血和血浆黏度;测定脑组织SOD活性、MDA含量;检测血浆血栓素A2(TXA2)和前列环素(PGI2)的含量。结果表明,假手术组大鼠神经行为学评分、组织细胞形态无明显异常;模型组神经行为学评分显著升高,HE染色出现明显病理学改变,表现神经细胞及血管周围间隙加大,神经细胞及胶质细胞肿胀,胞核碎裂或消失,呈现明显的脑水肿改变,尤为海马回明显。血清丙二醛(MDA)及血栓素A2(TXA2)含量升高,超氧化物歧化酶(SOD)活性及前列环素(PGI2)水平降低;蛭龙通络片中、大剂量可明显减轻大脑中动脉缺血再灌注模型大鼠的神经症状,降低神经生物学评分,降低脑含水量及脑指数,明显减轻脑神经细胞病理学改变,明显降低血清MDA及TXA2含量,提高SOD活性及PGI2水平,亦能明显降低全血黏度及血浆黏度,上述作用与阳性对照药步长脑心通胶囊之间无明显差异。提示蛭龙通络片对大鼠大脑中动脉缺血再灌注损伤具有明显保护作用。
