核桃楸皮化学成分及酚氧化酶抑制活性的研究

摘要
核桃楸皮为胡桃属(Juglans)植物核桃楸(Juglans mandshurica Maxim.)的树皮。该植株树皮、果皮、枝叶和根部均可入药。胡桃属植物具有消除自由基、抗肿瘤、抗氧化及杀虫等作用。本课题就核桃楸皮的化学成分及其对酚氧化酶(PO)的抑制活性进行研究。预实验以石油醚、氯仿、正丁醇三个萃取层组分和水相残留物简单的进行PO活性测定,发现正丁醇层抑制作用最大,因此实验重点研究该相萃取物成分和活性。利用硅胶柱色谱、凝胶柱色谱和HPLC等分离技术分离纯化,根据其理化性质和波谱特征鉴定共得到21个化合物:7个黄酮类物质,槲皮素-3-O-α-L-鼠李糖苷(8)、山奈酚-3-O-a-L-鼠李糖苷(9)、二氢槲皮素(11)、山奈酚(15)、大豆异黄酮(18)、二氢山奈酚(20)、柚皮素(21);8个酚酸类,3,3'-二甲氧基鞣花酸(1)、鞣花酸(4)、4'-二氢红花菜豆酸(12)、没食子酸(13)、3,3'-dimethoxyellagic acid-4'-O-P-D-xylose (14)、没食子酸甲酯(16)、没食子酸乙酯(17)、4,4'-dimethoxyellagic acid (19);3个二芳基庚烷,rhoiptelol (6)、(3R)-3',4"-epoxy-1-(4-hydroxyphenyl)-7-(3-methoxyphenyl) heptan-3-ol (7)、rhoiptelol C (10); juglanthrthracenoside A(2)、核桃酮(3)及p-谷甾醇(5),其中物质12、14、16、17、18、19为首次从胡桃属中获得。选取分离出的黄酮、鞣花酸、二芳基庚烷三类物质研究了它们对PO的抑制活性。结果发现上述三类物质均对PO有抑制作用,其中rhoiptelol C (10), dihydrokaempferol (20)和naringenin(21)三个化合物抑制作用较强,它们IC50是熊果苷的5-10倍。由于PO抑制剂在医农药、食品、化妆等领域有着广泛的应用,研究意义较大。
