坛紫菜和龙须菜多糖对巨噬细胞免疫调节作用的研究

摘要
巨噬细胞是广泛分布于机体的一种重要的免疫细胞,主要起源于骨髓前体细胞。巨噬细胞能被外界刺激激活,被活化的巨噬细胞其吞噬能力显著增强,能够通过直接吞噬或者分泌炎性细胞因子(IL-6、TNF-α、IL-1β等)、NO和活性氧等抗病原介质间接杀死病原体,有效防御外源病原体对机体的侵害,是机体的第一道防线。巨噬细胞还能够通过吞噬病原体并呈递抗原,从而诱发机体的免疫应答来抵御外界的侵害。海藻多糖近年来被发现具有多种生物活性,如抗肿瘤、抗氧化、抗病毒、降血脂、免疫调节等。海藻资源丰富,探究其生物活性成分,开发功能性食品从而达到高值化利用具有重要意义。本研究通过水煮醇沉、DEAE-cellulose纤维柱层析获得较高纯度的紫菜多糖和龙须菜多糖;紫菜多糖的糖含量为81.20%,糖醛酸含量为7.35%,硫酸根含量为14.67%;龙须菜多糖的糖含量为81.19%,糖醛酸含量为8.20%,硫酸根含量为9.33%。利用巨噬细胞RAW264.7探究紫菜多糖和龙须菜多糖的生物活性。MTT实验显示,两种多糖都能够有效促进RAW264.7的增殖,增强细胞活性;在5~100μg/m L浓度范围内,紫菜多糖刺激12 h,其促增殖作用随浓度升高而升高,刺激24 h,浓度在40μg/m L时细胞活力达到最高,是对照组细胞活力的2.7倍;龙须菜多糖浓度在12.5~200μg/m L范围内能够显著增强RAW264.7细胞活力,但各浓度之间无显著差异。中性红摄取实验显示,紫菜多糖和龙须菜多糖能够显著增强巨噬细胞的吞噬能力,不同浓度(25、50、100μg/m L)的紫菜多糖处理组RAW264.7吞噬能力分别是对照组的1.8、2.1、2.5倍,不同浓度(50、100、200μg/m L)的龙须菜多糖处理组RAW264.7吞噬能力分别是对照组的1.7、2.0、2.4倍。此外,紫菜多糖和龙须菜多糖都能够有效促进RAW264.7释放NO、TNF-α、IL-6等细胞因子;RAW264.7经不同浓度(25、50、100μg/m L)紫菜多糖刺激后,NO释放量为34.76、49.20、53.18μmol/L,与对照组(1.722μmol/L)相比显著上升,TNF-α含量为136.76、147.08、141.72 ng/L,与对照组(109.84 ng/L)相比显著上升,IL-6的含量(147.17、160.55、155.13 ng/L)与对照组(133.01 ng/L)相比显著上升;经50、100、200μg/m L龙须菜多糖刺激后,NO释放量由对照组0.21μmol/L上升到50.31、55.13、69.66μmol/L,TNF-α含量为132.20、141.27、149.93ng/L,与对照组(107.34 ng/L)相比显著上升,IL-6的含量(142.20、151.27、159.93 ng/L)与对照组(127.34 ng/L)相比显著上升。实验结果表明,紫菜多糖和龙须菜多糖促进RAW264.7释放NO主要是通过JNK和JAK2两条信号通路途径。综上所述,从坛紫菜和龙须菜中纯化的多糖能够有效激活RAW264.7,促进其细胞的增殖,增强其吞噬能力,并调节RAW264.7分泌相关细胞因子。
