降钙素对软骨细胞炎症反应及p38MAPK信号通路的影响

摘要
目的本实验通过观察降钙素对白介素1β(IL-1β)诱导的大鼠软骨细胞中基质金属蛋白酶13(MMP-13)、Ⅱ型胶原(ColⅡ)、p38、P-p38及c-myc表达的影响,探讨降钙素是否通过阻断p38MAPK信号通路抑制IL-1β引起的大鼠软骨细胞的炎性损伤。方法体外获得、培养大鼠关节软骨细胞。采用甲苯胺蓝及免疫细胞化学染色方法鉴定软骨细胞。MTT法检测细胞增殖率,确定实验所需降钙素浓度。将第2代软骨细胞分为7组,分别为:A组空白对照组,加入完全培养基;B组IL-1β诱导组,加入含10ng/ml IL-1β的完全培养基;C组降钙素对照组,加入含50ng/ml降钙素的完全培养基;D组IL-1β+降钙素干预组,加入含10ng/ml IL-1β的完全培养基15min后再加50ng/ml降钙素;E组IL-1β+SB203580干预组,加入含10ng/ml IL-1β的完全培养基15min后再加10μmol/L SB203580;F组降钙素+SB203580联合对照组,加入含50ng/ml降钙素和10μmol/L SB203580的完全培养基;G组IL-1β+降钙素+SB203580联合干预组,加入含10ng/ml IL-1β的完全培养基15min后再加入50ng/ml降钙素和10μmol/L SB203580。24h后做相应处理,用免疫细胞化学染色、Western Blot观察和检测MMP-13、ColⅡ、p38、P-p38及c-myc的表达情况。结果1甲苯胺蓝和ColⅡ免疫细胞化学染色结果说明体外培养细胞为软骨细胞,能够用来进行下一步实验;2 MTT结果提示浓度在50-500ng/ml范围的降钙素对软骨细胞抑制率无显著差异,故实验中的降钙素浓度选取50ng/ml浓度。3 Western Blot结果:1)各组p38表达无显著差异(P>0.05);P-p38在B组的表达显著高于其它各组(P<0.05),在D、E、G组的表达高于A组而低于B组(P<0.05),D、E、G组间无显著差异(P>0.05);2)c-myc和MMP-13在B组的表达显著高于其他各组(P<0.05),在D、E组的表达高于A组而低于B组(P<0.05),D、E组间无显著差异(P>0.05),在G组的表达低于B、D、E组而高于A组(P<0.05);3)ColⅡ在B组的表达显著低于其它各组(P<0.05),在D、E、G组的表达低于A组而高于B组(P<0.05),D、E、G组间无显著差异(P>0.05);各A、C、F组间无显著差异(P>0.05)。4对MMP-13、ColⅡ、p38、P-p38及c-myc进行免疫细胞化学染色,其结果支持Western Blot结果。结论IL-1β可以体外诱导大鼠软骨细胞发生炎性损伤。降钙素对IL-1β诱导的大鼠软骨细胞炎性损伤具有一定的抑制作用,其机制之一可能是通过抑制p38MAPK信号通路的激活,从而下调MMP-13的表达,上调ColⅡ的表达。
