脂多糖对体外诱导培养的小鼠破骨细胞分化和活性的影响

摘要
骨关节感染激活破骨细胞过度分化和活化,导致破骨细胞骨吸收与成骨细胞的骨形成失平衡,是形成骨溶解、骨不愈合的重要因素。脂多糖(LPS)是革兰氏阴性菌外胞膜的活性成分,通过直接和间接途径调节破骨细胞的分化。研究LPS介导下破骨细胞分化和活性调控机制对于明确感染性骨疾病中骨生长具有重要意义。 目的:研究脂多糖对体外诱导培养的破骨细胞分化和活性的影响。 方法:(1)采用不同浓度的脂多糖(lipopolysaccharide, LPS)对小鼠单核细胞RAW264.7细胞进行诱导培养。培养第4天进行抗酒石酸酸性磷酸酶(tartrate-resistant acidphosphatase, TRAP)染色计数;定量PCR检测破骨细胞TRAP、基质金属蛋白酶9(matrixmetalloproteinase-9, MMP-9)、组织蛋白酶K(cathepsin K, CK)等破骨相关基因表达;蛋白印迹试验检测核因子kB受体活化因子(receptor activator of NF-кB, RANK)、肿瘤坏死因子受体相关因子6(tumor necrosis factor receptor-associated factor6, TRAF6)及环氧合酶-2(COX-2)等蛋白表达。 (2)采用小鼠巨噬细胞集落刺激因子(macrophage-colony stimulating-factor,M-CSF)和小鼠重组可溶性核因子kB受体活化因子配基(receptor activator of NF-кBligand, RANKL)对小鼠单核细胞RAW264.7进行诱导培养,于培养第4天加入不同浓度LPS。培养第5天RT-PCR检测破骨细胞TRAP、MMP-9、RANK、CK等破骨相关基因及COX-2表达。 结果:(1)LPS单独诱导RAW264.7细胞,于培养第4d出现TRAP染色阳性多核破骨细胞。LPS上调破骨细胞相关基因TRAP、MMP-9、CK及蛋白RANK、TRAF6、COX-2的表达。尤以100ng/ml的LPS为著。 (2)LPS亦能够上调诱导的破骨细胞相关基因TRAP、MMP-9、 RANK、CK及COX-2.的表达。尤以100ng/ml的LPS为著。 结论:(1)LPS能刺激破骨前体细胞分化为破骨细胞 (2)LPS通过上调破骨相关基因表达促进已分化破骨细胞活性 (3)LPS是引起感染性骨疾病中骨溶融的重要因素
