龙眼体细胞胚胎发生机理的初步研究

本研究以龙眼“红核子”胚性愈伤组织为材料,对龙眼 体细胞胚胎发生进行同步化调控,采用石蜡切片、扫描电 镜、透射电镜观察,内源激素、多胺测定,同工酶电泳和 蛋白质IEF和SDS-PAGE等手段,探讨龙眼体细胞胚胎发 生的机理。 从龙眼体胚发生的石蜡切片、扫描电镜、透射电镜观 察结果发现,龙眼胚性愈伤组织的胚胎发生是内起源的, 并且起源于单细胞;细胞要表现出胚胎发生的潜力需与邻 近细胞保持相对独立;在体胚发育过程中超微结构变化明 显,特别是早期玻璃化子叶形胚细胞壁皱折、内含物少, 而经成熟培养后的成熟胚细胞形状规则、内含物丰富。 龙眼胚性愈伤组织中内源激素、多胺含量比非胚性愈 伤组织高。体胚发生过程中内源激素、多胺变化趋势大致 呈“M”状。在0.8mg/12,4-D培养基控制下的胚性愈伤组 织和球形胚为变化的2个明显转折点。内源激素和多胺的 变化先于形态学的变化。 非胚性愈伤组织同工酶谱带和蛋白质组分较少,活性 较弱,13.9KD的蛋白质为非胚性愈伤组织的特异蛋白质。 胚性愈伤组织出现了一些新的蛋白组分和同工酶谱带,从 胚性愈伤组织到球形胚阶段变化不大。玻璃化早期子叶形 胚出现pI 4.4的特异蛋白质。成熟子叶形胚出现了很特别 的、分子量为6.9KD、6.2KD的小分子量特异蛋白质,推测 它们在体细胞胚胎发育中起着重要的作用,并且可能与龙 眼这类具大核的木本植物的特性有关。
