FLUOROMETRIC ASSAY OF ALPHA-METHYLENE CARBONYL-COMPOUNDS WITH N1-METHYLNICOTINAMIDE CHLORIDE

摘要
A spectrophotofluorometric method has been developed for the determination of α-methylene carbonyl compounds by using N1-methylnicotinamide chloride (NMN). When reacted at 0 °C for 10 min with NMN in the presence of alkali and heated at 92 °C for 3 min after the addition of formic acid solution, only compounds having an α-methylene carbonyl group (-CH2-CO-) gave intense violet to green fluorescence. By measuring the fluorescence, as little as 5 pmol of ace-tophenone, 1-indanone and α-tetralon could be determined. The relative standard deviations of 1.00% and 1.61% are observed for the analyses of 1 X 10ˉ 9 mol and 1 X 10ˉ10 mol of acetophenone, respectively.
