A biological approach to the molecular nature of mutations induced by two alkylating agents (ethyl methanesulfonate and methyl methanesulfonate), nitrous acid and hydroxylamine is reported. Extensive phenotypic and genetic analyses have been performed on mutations induced in the ad-6 and ad-7 loci of the yeast Schizosaccharomyces pombe. Hydroxylamine, nitrous acid and ethyl methanesulfonate produced almost exclusively (95-99%) base-pair substitutions in DNA. Among the mutations induced by methyl methanesulfonate 15% of possibly insertion-deletion mutants was found. © 1966.
