ENHANCED FRACTION RELEASABLE ABOVE PERCOLATION-THRESHOLD FROM MONOLITHS CONTAINING OSMOTIC EXCIPIENTS

摘要
Ethylene-co-vinyl acetate monoliths containing osmotic excipients of varying particle sizes and at various volume fractions were prepared. The excipients chosen were potassium chloride, sodium chloride, sodium salicylate, sodium phosphate monobasic, sodium carbonate and bovine serum albumin. The total fraction releasable was found to increase as the volumetric loading and the excipient osmotic activity increased, and was weakly inversely dependent on particle size. The total fraction released was enhanced by osmotic pressure induced polymer rupturing of encapsulated particles. A model was developed, based on the concept of a distribution of particle wall thicknesses around encapsulated particles in the monolith, and the existence of a critical wall thickness for rupture, to explain the fraction releasable enhancement. The model effectively explains the contribution of osmotic rupturing to the overall fraction releasable.
