瑞舒伐他汀预处理对心肌缺血再灌注损伤大鼠心肌细胞自噬的影响附视频

摘要
目的探讨瑞舒伐他汀预处理对心肌缺血再灌注损伤(MIRI)大鼠心肌细胞自噬的调控作用。方法将40只Wistar大鼠随机分为假手术组、模型组、瑞舒伐他汀低剂量组、瑞舒伐他汀高剂量组。模型组、瑞舒伐他汀低剂量组、瑞舒伐他汀高剂量组制备大鼠MIRI模型,假手术组同样方法开胸,只穿线不结扎冠状动脉。模型建立前7天,瑞舒伐他汀低剂量组、瑞舒伐他汀高剂量组分别给予瑞舒伐他汀5、20 mg/kg灌胃一次,假手术组及模型组给予同体积生理盐水。观察各组大鼠左心室血流动力学参数[左心室舒张末压(LVEDP)、左心室内压最大上升和下降速率(±dp/dtmax)]改变; HE染色法观察大鼠心肌组织病理学改变; Western blotting法检测大鼠心肌细胞自噬相关因子LC3-Ⅱ、Beclin-1蛋白表达。结果模型组大鼠LVEDP、LC3-Ⅱ蛋白、Beclin-1蛋白表达水平高于假手术组(P均<0. 05),±dp/dtmax水平低于假手术组(P <0. 05);瑞舒伐他汀低剂量组、瑞舒伐他汀高剂量组大鼠LVEDP、LC3-Ⅱ蛋白、Beclin-1蛋白表达水平低于模型组(P均<0. 05),±dp/dtmax水平高于模型组(P <0. 05),且瑞舒伐他汀高剂量组较瑞舒伐他汀低剂量组改变更为显著(P均<0. 05)。结论瑞舒伐他汀预处理可减轻大鼠MIRI引起的心肌组织损伤,这可能与抑制心肌细胞自噬有关。
