摘要
<正> 限制性内切酶(restriction endonuclease,简称限制酶)是一类能识别并切开特定DNA序列的核酸内切酶。已发现的限制酶将近500种,其中50余种已广泛应用。限制酶分为三型:Ⅰ型酶由不同的亚基组成,具有ATP依赖性的水解活性和甲基化活性。Ⅱ型酶由相同亚基构成,仅具限制酶活性,它们能识别的DNA专一性序列通常为4~6个碱基对,并在特定位点将底物水解,因而在分子遗传学中广泛应用。Ⅲ型酶仅有数种,如HinfⅢ、EcoPⅠ等,但其切点未定。
