基因的表达同时受到生理信号和环境刺激的调节。近年来,活性氧被认为是多种生长因子和细胞因子信号通路中的一种第二信使,同时其也产生在细胞损伤应激过程中。活性氧对转录因子的半胱氨酸位点的氧化还原状态有重要的决定作用,其主要影响转录因子二聚体构象的形成、与DNA结合的关键半胱氨酸位点的氧化还原状态、锌指等蛋白质构象的形成。关于氧化还原环境对控制增殖、分化、凋亡功能的转录因子OxyR、Yap-1、AP1、NF-κB、p53、Sp1、USF、HIF-1、C/EBP等的调节,已有大量研究提示氧化还原调节是一种关键的基因表达的调节方式。本文总结了氧化修饰对转录因子发挥调节作用的一般原理和若干典型例子。
