丹参酮ⅡA对缺血性心力衰竭大鼠心肌纤维化的影响

摘要
目的研究丹参酮ⅡA通过抑制Galectin-3表达对缺血性心力衰竭大鼠心肌纤维化的影响。方法 36只Wistar大鼠随机均分为3组:假手术组(Sham),缺血性心力衰竭组(IHF),丹参酮ⅡA组(TSA),建立缺血性心力衰竭模型,其中丹参酮ⅡA组于结扎冠脉前1周每天给予TSA预处理,剂量为40mg/kg,通过心脏彩超观察手术后4周各组心功能情况,通过Masson染色观察各组大鼠心肌纤维化程度,蛋白免疫印迹法测定Galectin-3蛋白表达情况。结果 TSA组与IHF组相比,心功能参数均出现显著改善(P均<0. 01); Masson染色示,TSA组明显降低了缺血性心力衰竭所造成的大片胶原纤维增生,深蓝色显色较少,心肌纤维走形平整。Western blot法结果显示,与IHF组比较,TSA组显著降低了由缺血性心力衰竭所造成的Galectin-3的上升(0. 52±0. 09 vs 1. 43±0. 18,P <0. 01)。结论丹参酮ⅡA可有效抑制缺血性心力衰竭大鼠心肌纤维化,其作用可能与抑制Galectin-3表达相关。
