基于全病毒颗粒的中东呼吸综合征冠状病毒抗体检测方法的建立

摘要
目的建立基于全病毒颗粒的中东呼吸综合征冠状病毒(Middle East respiratory syndrome coronavirus, MERS-CoV)感染的抗体检测方法, 并将该方法用于MERS相关病例血清抗体(IgG与IgM)的检测。方法采用基于灭活MERS-CoV颗粒作为检测用靶抗原, 建立酶联免疫检测方法(ELISA), 选取10份新生儿血清和40份正常成年人血清标本, 确定MERS-CoV IgM和IgG抗体检测界值;在此基础上对我国第1例MERS感染病例的5份系列血清标本抗MERS-CoV抗体进行检测。结果全病毒灭活颗粒抗原与正常成人或儿童血清不存在明显交叉反应, 可确定MERS-CoV抗体IgM和IgG检测界值A450分别为0.32和0.42。我国输入性MERS病例入院初期血清IgM和IgG抗体滴度均为1︰40, 入院2周后IgM和IgG抗体阳转, 滴度均有4倍以上升高, 入院4周后IgG滴度可达到1︰640。结论建立了基于灭活全病毒颗粒的MERS-CoV特异性抗体的ELISA检测方法, 该方法可用于MERS病例血清抗体(IgG与IgM)的检测。
