血红素加氧酶-1调控动脉粥样硬化进程和CD4+T细胞分化的研究

摘要
目的探讨血红素加氧酶-1(HO)-1对Apo E基因敲除(Apo E-/-)小鼠动脉粥样硬化(AS)进程和效应性CD4+T细胞极性分化的影响。方法将8周龄雄性Apo E-/-小鼠随机分为两组,给予8周西方饮食(含0.15%胆固醇饲料)的同时,分别经腹腔注射生理盐水(对照组)和Hemin(诱导HO-1,HO-1组),并维持8周。分光光度法检测HO-1活性,流式细胞术检测CD4+T细胞比例,ELISA检测血浆脂质和细胞因子水平。结果与对照组相比,HO-1组动脉内HO-1表达以及HO-1活性明显增加,血浆脂质氢过氧化物浓度降低,斑块面积显著减轻,小鼠脾脏Th1、Th2和Th17比例显著降低,血浆IFN-γ、IL-4和IL-17表达下降,差异有统计学意义(P<0.05,P<0.01,P<0.001)。结论 HO-1通过调控效应性T细胞极性变化抑制AS进程。
