BADGE联合锂盐通过抑制PPARγ预防家兔激素性股骨头坏死

摘要
目的:研究双酚A二缩水甘油醚(BADGE)单独或与锂盐结合治疗对家兔骨髓间充质干细胞成脂分化和激素性股骨头坏死(ONFH)的预防作用及其可能机制。方法:分离家兔骨髓间充质干细胞并诱导成脂分化,油红O染色验证BADGE联合锂盐对成脂分化的抑制作用。将兔随机分为4组,除对照组外,将甲强龙(MPS)造模的家兔分为3组:一组行BADGE联合锂盐(BL)治疗,另一组行BADGE单独治疗(B),最后一组行空白治疗(M)。2周采血检测血脂,4周后提取双侧股骨头做病理、μ-CT、荧光定量PCR检测。结果:BADGE处理抑制了兔骨髓间充质干细胞(BMSC)成脂分化,并抑制脂肪分化主要调节因子PPARγ和C/EBPα的表达,并且这些生物活性与锂盐结合得到进一步增强。在体内研究中,结果显示BADGE治疗可降低激素性ONFH发生率,减少骨髓肥胖,并且改善了骨密度及骨质量,而且这些效果在与锂盐联合得到增强。BADGE单独治疗或与锂盐结合增加了RUNX2和β-catenin的表达,并抑制了PPARγ和C/EBPα的表达,同时降低了血清总胆固醇(TC)和游离脂肪酸(FFA)水平。结论:通过抑制PPARγ,BADGE单独或与锂盐联合治疗有效预防了激素性股骨头坏死。
