牙本质非胶原蛋白对大鼠骨髓间充质干细胞增殖活性和矿化能力的影响

摘要
目的:探讨EDTA提取的牙本质非胶原蛋白(DNCP)对体外培养的大鼠骨髓间充质干细胞生物学性能的影响。方法:通过MTT法、碱性磷酸酶活性检测、茜素红染色和氯代十六烷基吡啶定量检测法观察EDTA提取的DNCP对大鼠骨髓间充质干细胞的增殖活性和矿化能力的影响。结果:经DNCP诱导1、3、5、7、9d,各浓度组大鼠骨髓间充质干细胞增殖活性与对照组无差别(P>0.05);诱导后5d和7d时,10μg/mL组和1μg/mL组均可上调大鼠骨髓间充质干细胞ALP活性,与对照组相比有统计学差异(P<0.05);诱导3周后,茜素红染色显示10μg/mL组出现较大的红色结节,定量分析显示,其形成钙化结节的能力明显高于对照组(P<0.05)。结论:DNCP对大鼠骨髓间充质干细胞的增殖活性影响不明显;高浓度DNCP可促进大鼠骨髓间充质干细胞的ALP活性和矿化形成能力。
