红车轴中异黄酮类物质的分析鉴定及活性评价研究

摘要
目的采用高效液相色谱-电喷雾质谱联用技术(LC-ESI-MS)对红车轴乙醇提取物中的化学成分进行分析鉴定,筛选红车轴中α-葡萄糖苷酶及黄嘌呤氧化酶抑制剂。方法选用反相C18色谱柱,流动相A为0.05%磷酸水溶液,流动相B为乙腈,流动相梯度程序:0~20 min,10%55%B;流速0.8ml/min;样品进样量为10μl;检测波长:260 nm,利用电喷雾质谱(ESI-MS)负离子模式在线检测化学成分;以α-葡萄糖苷酶及黄嘌呤氧化酶作为生物靶分子,以超滤质谱技术筛选酶抑制剂。结果共鉴定出5个化学成分,分别为大豆素(daidzein),樱黄素(prunetin),染料木素(genistein),得鸢尾素(irilone)和山槐素(maackiain)。该5个化合物均可与黄嘌呤氧化酶亲和,除Gartanin外均可以与α-葡萄糖苷酶亲和。结论利用液质联用技术对红车轴中化学成分进行分析可以快速分析鉴定红车轴中的化合物并提供结构信息。利用超滤质谱技术筛选有效成分具有快速和灵敏等优势。
