生长分化因子15对过氧化氢诱导的人主动脉内皮细胞凋亡的保护作用

摘要
目的研究生长分化因子15(GDF-15)对过氧化氢(H2O2)诱导的人主动脉内皮细胞凋亡的保护作用及其可能的作用机制。方法体外培养人主动脉内皮细胞,随机分为空白对照(A)组、H2O2损伤(B)组、低浓度GDF-15+H2O2(C)组、高浓度GDF-15+H2O2(D)组、低浓度GDF-15+H2O2+LY294002(E)组和高浓度GDF-15+H2O2+LY294002(F)组。采用Hoechst3258染色、流式细胞术检测细胞凋亡情况,Western blot检测磷酸化Akt(p-Akt)蛋白相对表达量。结果 B组细胞凋亡率高于A组(P<0.05)。与B组相比,C组、D组细胞凋亡率降低,p-Akt蛋白表达增加,并呈剂量依赖性(P<0.05);而经LY294002预处理后,E组、F组细胞凋亡率增高,p-Akt蛋白表达减少(P<0.05)。结论 GDF-15可拮抗H2O2诱导的内皮细胞凋亡。其作用机制可能与上调p-Akt的表达有关。
