防己诺林碱通过Nrf2/NLRP3途径对去卵巢骨质疏松大鼠骨结构和成骨细胞凋亡的影响

摘要
目的探讨防己诺林碱通过Nrf2/NLRP3途径对去卵巢骨质疏松大鼠骨结构和成骨细胞凋亡的影响及机制研究。方法 SD大鼠分为假手术组(sham组)、骨质疏松症组(OP组)、防己诺林碱低剂量组(OP+FAN-L组)和防己诺林碱高剂量组(OP+FAN-H组)。μCT检测各组大鼠骨组织的骨体积分数(BV/TV)、骨小梁厚度(Tb.Th)和骨小梁分离距离(Tb.Sp);ELISA检测各组大鼠血清中碱性磷酸酶(ALP)和骨钙素(OC)的表达水平;ELISA检测各组大鼠骨组织中白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的表达水平;HE染色检测各组大鼠骨组织病理损伤;TUNEL染色检测各组大鼠成骨细胞的凋亡;Western blot检测各组大鼠骨组织中核因子E2相关因子2(Nrf2)和核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)蛋白的表达。结果防己诺林碱使OP大鼠的BV/TV和Tb.Th指数上升(P<0.05),Tb.Sp指数下降(P<0.05);与OP组相比,OP+FAN-L组和OP+FAN-H组大鼠血清中ALP的表达水平下调(P<0.05),OC的表达水平上调(P<0.05),骨组织中IL-6和TNF-α的表达水平下调(P<0.05);防己诺林碱使OP大鼠骨组织中骨小梁面积增加(P<0.05);与OP组相比,OP+FAN-L组和OP+FAN-H组TUNEL阳性细胞数量减少(P<0.05),骨组织中Nrf2蛋白表达水平上调(P<0.05),NLRP3蛋白表达水平下调(P<0.05)。结论防己诺林碱减少了炎症因子的表达和成骨细胞凋亡,增加了骨组织中骨小梁面积,其可能与上调Nrf2的表达和抑制NLRP3的表达有关。
