RP-HPLC同时测定黄连解毒汤7种活性成分

摘要
目的建立一种同时测定黄连解毒汤中7种活性成分(栀子苷、黄芩苷、盐酸巴马汀、盐酸小檗碱、汉黄芩苷、黄芩素和汉黄芩素)含量的RP-HPLC方法。方法采用迪马Diamonsil C18色谱柱(250×4.6 mm,5μm),以乙腈(A)-0.05%磷酸二氢钾和0.05%三乙醇胺(用磷酸调pH到3.0)(B)为流动相,梯度洗脱:0~9 min,84%B;9~27 min,84%→73%B;27~35 min,73%→70%B;35~39 min,70%→68%B;39~42 min,68%→60%B;42~46 min,60%→52%B;46~53 min,52%→45%B;53~60 min,45%→84%B。流速0.8 ml/min,检测波长254 nm,柱温35℃。结果在60 min内黄连解毒汤中栀子苷、黄芩苷、盐酸巴马汀、盐酸小檗碱、汉黄芩苷、黄芩素和汉黄芩素7种成分被完全分离;回归方程显示7种成分的峰面积与其浓度呈良好的线性相关;7种成分的加样回收率(n=6)为94.92%~106.76%,RSD均小于2.0%。结论利用反相高效液相色谱法,建立了一种简便准确,可同时测定黄连解毒汤中7种活性成分含量的检测方法。
