电针“长强”穴对FMR1基因敲除小鼠海马CA1区NLGN-3蛋白表达的影响

摘要
目的:探讨电针"长强"穴对FMR1基因(fragile X mental retardation 1,脆性X智力迟钝蛋白1)敲除小鼠学习记忆能力及海马CA1区NLGN-3蛋白(neuroligin-3,神经连接蛋白-3)表达的影响。方法:(1)将24只23日龄FMR1基因敲除小鼠随机分为长强组、非穴组、模型组,每组8只,长强组采取电针干预,波型为连续波,频率为2 Hz,强度为2m A,持续20 min,1周6次,干预2周;非穴组以小鼠右助弓最低点上1 cm处作为非经非穴点,电针操作与长强组一致;模型组每日只进行同样的抓取固定,不采取任何干预。(2)所有分组于23日龄及41日龄进行Morris水迷宫检测小鼠干预前后的学习记忆能力,于46日龄进行免疫组化法检测小鼠海马CA1区NLGN-3蛋白的表达。(3)统计方法:SPSS20.0统计软件对实验数据进行单因素方差分析。结果:对比模型组、非穴组,干预后长强组逃避潜伏期降低(P<0.05);原平台象限游泳路程/总路程比值增高(P<0.05),海马CA1区NLGN-3蛋白表达增高(P<0.05)。结论:电针"长强"穴能提高FMR1基因敲除小鼠的学习记忆能力,其机制可能与调控海马CA1区NLGN-3蛋白表达有关。
