高脂血症对脑缺血大鼠血管调节因子的影响

摘要
目的比较高脂血症条件下复合脑缺血大鼠模型在缺血不同时间血管内皮细胞功能及血管调节因子的变化特点,分析高血脂因素对脑缺血性损伤的影响。方法实验动物随机分为正常组、高脂血症组、假手术组、高脂假手术组、单纯脑缺血组、复合脑缺血组,高脂饲料喂养大鼠,制备高脂血症大鼠模型线栓法建立大鼠大脑中动脉缺血模型(MCAO)。分别在缺血3天、7天取材,用酶联免疫法(ELISA)检测血清中血管性血友病因子(vWF)、一氧化氮(NO)、内皮素1(ET-1)、6-酮-前列腺F1a(6-keto-FGF1α)(前列环素PGI2的代谢产物)、血栓素B2(TXB2)(血栓素TXA2的代谢物)的含量。结果与相应假手术组相比较,复合脑缺血组和单纯脑缺血组中vWF和TXB2因子在脑缺血模型各时间点血清含量呈增多趋势;vWF在单纯脑缺血7天和复合脑缺血3天血清含量均增多,具有统计学意义(P<0.05);而ET-1的含量无明显变化(P>0.05);TXB2单纯脑缺血3天和7天血清含量均增多,具有统计学意义(P<0.05),而在复合脑缺血的血清中含量无明显变化(P>0.05);NO、6-keto-FGF1α在缺血3天、7天血清含量均高于相应的假手术组,差异具有统计学意义(P<0.05)。复合脑缺血组与单纯脑缺血组组间比较,vWF复合脑缺血组在缺血3天、7天血清含量均增多(P<0.05);ET-1的含量无显著性变化(P>0.05);TXB2复合脑缺血组血清含量减少,缺血3天、7天含量均明显增加(P<0.05);NO、6-keto-FGF1α复合脑缺血组在缺血3天、7天血清含量均减少,但在缺血3天组间比较无明显变化(P>0.05),缺血7天组间明显增加(P<0.05)。结论高脂血症条件下,血管调节因子在缺血不同时期既有累积现象,又有特异性的表达,可能在脑缺血恢复期有助于损伤的修复。
