银杏二萜内酯葡胺注射液对缺糖缺氧损伤的SH-SY5Y细胞保护作用

摘要
目的探讨银杏二萜内酯葡胺注射液(YXETNZ)对缺糖缺氧(oxygen-glucose deprivation,OGD)损伤的人神经母细胞瘤细胞(SH-SY5Y)的保护作用及可能的机制。方法 SHSY5Y细胞OGD损伤4 h后,与药物一起复氧1 h,然后测定细胞存活率(CCK-8法)、caspase-3/7酶活力、细胞质中核小体含量;蛋白质免疫印迹检测细胞中p-Akt、p-PKA、p-Bad蛋白量的变化。结果 OGD明显提高SH-SY5Y细胞caspase-3/7酶活力和细胞质中核小体含量,明显降低细胞的存活率。YXETNZ能明显提高OGD损伤的SH-SY5Y细胞的存活率,抑制caspase-3/7酶活性,减少细胞核核小体的释放量,上调p-Akt、p-PKA和p-Bad激酶蛋白量,提高Akt和PKA活性,保护神经细胞。结论 YXETNZ对OGD损伤的SHSY5Y细胞具有明显的保护作用,其保护机制可能与细胞内PI3K/Akt/Bad/caspase-3/7、c AMP/PKA/Bad/caspase-3/7细胞通路的激活有关。
