乳香-没药治疗癌性疼痛的网络药理学研究

摘要
目的采用网络药理学方法研究乳香-没药缓解癌性疼痛的分子靶点及通路。方法应用中药系统药理学数据库与分析平台(TCMSP)筛选药物主要成分,Uniprot网站获取药物化学成分相对应的人类相关基因,Genecards网站获取癌性疼痛相关基因,绘制韦恩图寻找共表达基因,利用STRING数据平台进行蛋白相互作用网络(PPI)分析,使用软件Cytoscape3.7.2构建成分-靶点相互作用图。利用基因本体(GO)、京都基因和基因组百科全书(KEGG)数据库对核心基因进行富集分析。结果获取乳香-没药有效成分53个,对应人相关基因92个,癌性疼痛相关基因8 585个。通过调控DNA盲转录激活活性、RNA聚合酶Ⅱ特异性等分子功能或过程,调节PI3K-Akt信号通路、MAPK通路来缓解疼痛。结论乳香-没药通过多种化学成分作用于多个靶基因有效缓解癌性疼痛,具体作用有待进一步实验验证。
