兴奋性氨基酸及其受体与转运体在抗癫痫药理学研究中的意义

摘要
目的:综述兴奋性氨基酸(EAA)、兴奋性氨基酸受体(EAARs)及兴奋性氨基酸转运体(EAATs)在抗癫痫药理研究中的意义。方法:以"Excitatory amino acids""Receptor""Transporter""Epilepsy"等为关键词,在Pub Med数据库中检索2004-2014年的相关文献,归纳总结后,从EAA、EAARs、EAATs与癫痫发生发展的关系以及在抗癫痫药理研究中的意义进行综述。结果与结论:检索到相关文献150余条,其中有效文献40条。中枢神经系统EAA水平异常升高、EAATs低表达都可导致谷氨酸(Glu)含量增加,从而诱发癫痫;EAARs因受到不适当刺激而产生兴奋毒性,导致神经元细胞损伤或缺失也是癫痫发作原因之一。EAA和代谢型Glu受体(m Glu Rs)可能对癫痫的治疗提供新的思路并为研制理想治疗药物提供了有益的靶标。作用于m Glu Rs和EAATs的药物、调节第二类m Glu Rs和EAATs的基因或影响相应蛋白表达的药物应用于临床,将对癫痫的防治发挥积极作用。
