五羟色胺2A受体基因-环境交互作用与抑郁伴自杀关联研究

摘要
目的探讨五羟色胺2A受体(5-hydroxytrytamine receptor 2A, 5-HTR2A)基因多态性、负性生活事件交互作用与抑郁伴自杀的关系。方法提取中国北方地区130名抑郁伴自杀者(病例组)和118名抑郁无自杀者(对照组)基因组DNA, 采用Snapshot技术分别扩增rs6311和rs6313位点的基因组DNA片段, 将PCR产物直接测序检测病例组和对照组的基因型, 应用二分类Logistic回归模型分析rs6311、rs6313与负性生活事件的交互作用。结果病例组与对照组rs6311位点的基因型(χ2=0.012)和等位基因频率(χ2=0.062)比较均差异无统计学意义(P>0.05)。两组rs6313位点的基因型比较, 差异无统计学意义(χ2=1.035, P>0.05), 但等位基因频率比较差异有统计学意义(等位基因A: 73.1%vs 50.4%;等位基因G:26.9%vs 49.6%;χ2=27.029, P<0.05)。rs6311和负性生活事件交互作用差异有统计学意义(OR=6.155, 95%CI=1.372~27.612, P<0.05)。rs6313和负性生活事件交互作用差异无统计学意义(OR=0.772, 95%CI=0.231~2.579, P>0.05)。结论 rs6311与负性生活事件的交互作用在抑郁伴自杀的发病机制中具有重要意义。
