双参芎连颗粒对ApoE基因敲除小鼠动脉粥样硬化炎症反应的干预作用

摘要
目的:研究双参芎连颗粒对高脂饲料喂养的Apo E基因敲除小鼠动脉粥样硬化炎症反应的干预作用。方法:采用C57BL/6小鼠做为正常组(BLA),ApoE-/-小鼠随机分为4组:动脉粥样硬化模型组(MOD),普罗布考治疗组(PRO,216 mg·kg-1),双参芎连颗粒高剂量治疗组(SSXL-H,1.6 g·kg-1)以及双参芎连颗粒低剂量治疗组(SSXL-L,0.8 g·kg-1)。各组ApoE-/-小鼠均给予高脂饲料喂养2周后采用生化法进行血脂测定,并灌胃相应的药物。给药8周后再次进行血脂检测。同时采用超声法检测主动脉弓内膜厚度;采用油红O染色的方法分析主动脉斑块面积进行计算斑块面积/血管内表面积;采用流式高通量多因子检测技术检测血清中肿瘤坏死因子(tumor necrosis factor,TNF)-α,白细胞介素(interleukin,IL)-1α,IL-10,以及单核细胞趋化蛋白(monocyte chemoattractant protein,MCP)-1等炎症细胞因子的表达。结果:双参芎连颗粒能够减少ApoE-/-小鼠主动脉中-内膜厚度,降低主动脉斑块面积/血管内表面积,抑制TNF-α和MCP-1炎症因子表达,但是对血脂水平无明显的影响。结论:双参芎连颗粒可以通过对炎症反应的抑制作用而防治动脉粥样硬化,且不依赖与血脂水平的变化。
