杨桃根提取物对CCl4致小鼠急性肝损伤的预防作用

摘要
目的探讨杨桃根提物(EACR)对CCl4致小鼠急性肝损伤的影响。方法预防性给予相应药物7 d后,复制急性肝损伤模型。检测血清中的谷草转氨酶(AST)、丙氨酸氨基转移酶(ALT)、白细胞介素1(IL-1)、白细胞介素6(IL-6)的水平,以及肝脏组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)的活性;Western blot检测肝脏组织中肿瘤坏死因子α(TNF-α)、核转录因子-κB(NF-κB)、caspase-3的蛋白表达水平;HE染色观察小鼠肝脏的病变情况。结果与模型组相比,给药组小鼠血清中AST、ALT、IL-1、IL-6及肝脏中MDA水平明显降低;肝脏组织中SOD、GSH、GSH-Px明显升高;肝脏组织TNF-α、NF-κB、caspase-3蛋白表达明显减少;HE染色显示肝脏组织病变减轻。结论 EACR通过降低酶活性,抑制脂质过氧化及炎症因子释放,达到保护小鼠肝脏的作用。
