黄芩汤治疗溃疡性结肠炎分子作用机制的网络药理学研究

摘要
目的基于网络药理学探讨黄芩汤治疗溃疡性结肠炎(Ulcerative colitis,UC)的分子作用机制。方法通过TCMSP数据库筛选黄芩汤的潜在活性成分(OB≥30%,DL≥0.18),采用Swiss数据库预测其作用靶点;与Disgenet数据库获得的UC疾病靶点相映射,得到黄芩汤治疗UC的潜在靶点。利用Cytoscape软件构建"黄芩汤-活性化合物-潜在作用靶点"网络和蛋白质相互作用网络,并使用David数据库对黄芩汤的作用靶点进行GO生物功能分析和KEGG通路富集分析。结果得到黄芩汤治疗UC关键靶点58个;甘草苷(Liquiritin)、甘草吡喃香豆酮(Licopyranocoumarin)、黄芩黄素(Baicalein)、黄连碱(Coptisine)、汉黄芩素(Wogonin)、鲨烯(Supraene)等可能是黄芩汤治疗UC的主要活性成分;其作用于ESR1、PTGS1、PTGS2、PTPN2、ABCG2等靶点,参与转录调控、低氧反应以及药物反应等生物过程,通过Ras-PI3K-Akt通路、Ras-PI3K-Akt-HIF-1α通路、NF-κB通路及MAPK-PI3K/AKT通路等发挥治疗UC的作用。结论黄酮类化合物在黄芩汤抗UC的作用机制中具有重要意义,调节炎症通路及干预炎症的发生发展可能是黄芩汤抗UC作用的重要机制。
