远程缺血后适应对脑缺血再灌注大鼠血脑屏障的保护作用

摘要
【目的】研究在大鼠脑缺血再灌注(IR)损伤模型中远程缺血后适应对血脑屏障的作用及相关机制。【方法】将SD大鼠96只随机分为3组:假手术组、单纯缺血再灌注组和远程缺血后适应组。利用线栓法制作大鼠脑IR模型,缺血1.5 h后恢复再通,再灌注72 h后对各组大鼠进行神经功能评分,染色检测脑梗死体积、梗死侧大脑半球脑组织伊文思蓝(EB)渗出量,再对紧密连接相关蛋白Claudin-5和Occludin的表达水平进行分析。【结果】与假手术组相比,单纯缺血再灌注组神经功能评分(9.9±2.0)、脑梗死体积(58.8±1.4)及EB的渗出量(5.7±0.6)均增加(P<0.01),同时Claudin-5和Occludin表达下降(P<0.01);与单纯缺血再灌注组比较,远程缺血后适应组大鼠神经功能评分(7.5±1.7)降低(P<0.01),脑梗死体积(33±5)减少(P<0.01),EB渗出量(2.7±1.2)减少(P<0.01),相应地Claudin-5和Occludin表达增加(P<0.01)。【结论】远程缺血后适应可以减轻脑缺血再灌注损伤,维持血脑屏障稳定性,可能与增加Claudin-5、Occludin的表达有关。
